Background

PC-766B is a macrolide antibiotic existed in anactinomycete strain SC-4710.

PC-766B was active against Gram-positive bacteria, and some fungi and yeasts, yet inactive against Gram-negative bacteria. PC-766B exhibited potent cytotoxicity against murine tumor cell lines in vitro. The IC50 values against P388 leukemia and B16 melanoma were 0.1 and 0.5 ng/ml, respectively [1]. Intraperitoneal administration of PC-766B (0.25mg/kg/day) showed in vivo antitumor activity against P388 leukemia. PC-766B showed a weak inhibitory activity against Na+, K+-ATPase [1].

General tips We do not recommend long-term storage for the solution, please use it up soon.

Protocol

Cell experiment [1]:

Cell lines

Murine cell lines P388 leukemia and B16 melanoma

Preparation method

The solubility of this compound in DMSO is soluble. General tips for obtaining a higher concentration: Please warm the tube at 37℃ for 10 minutes and/or shake it in the ultrasonic bath for a while. Stock solution can be stored below -20℃ for several months.

Reacting condition

0.1 ng/ml and 0.5 ng/ml

Applications

PC-766B showed strong cytotoxicity to murine tumor cell lines in vitro. IC50 values for P388 leukemia and B16 melanoma cell lines were 0.1 ng/ml and 0.5 ng/ml, respectively.

Animal experiment [1]:

Animal models

Male CDF1 mice

Dosage form

0.25 mg/kg (i.p.)

Application

Intraperitoneal injection of PC-766B (0.25mg/kg/day) showed antitumor activity against P388 leukemia in vivo.

Other notes

Please test the solubility of all compounds indoor, and the actual solubility may slightly differ with the theoretical value. This is caused by an experimental system error and it is normal.
